چکیده

Suboptimal cellular DNA repair capacity (DRC) has been shown to be associatedwith enhanced cancer risk, but genetic variants affecting the DRC phenotype have not been comprehensively investigated. In this study, with the available DRC phenotype data, we analyzed correlations between the DRC phenotype and genotypes detected by the Illumina 317K platform in 1,774 individuals of European ancestry from a Texas lung cancer genome-wide association study. The discovery phase was followed by a replication in an independent set of 1,374 cases and controls of European ancestry. We applied a generalized linear model with single nucleotide polymorphisms as predictors andDRC (a continuous variable) as the outcome. Covariates of age, sex, pack-years of smoking, DRC assay-related variables, and case–control status of the study participants were adjusted in the model. We validated that reduced DRC was associated with an increased risk of lung cancer in both independent datasets. Several suggestive loci that contributed to theDRCphenotypewere defined inERCC2/XPD, PHACTR2, andDUSP1. In summary, we determined that DRC is an independent risk factor for lung cancer, andwe defined several genetic loci contributing to DRC phenotype. Cancer Res; 73(1); 256–64. 2012 AACR.

No association between the XPD (Lys751G1n) polymorphism or the XRCC3 (Thr241Met) polymorphism and lung cancer risk.

Introduction DNA repair helps to protect the genome from tobacco-induced damage. Reduced DNA repair capacity may predispose individuals to lung cancer (1). Polymorphisms in genes encoding DNA repair proteins may modulate susceptibility to tobacco lung carcinogens. Bulky DNA adducts induced by chemical carcinogens in cigarette smoke are repaired through the nucleotide excision repair pathway tha...

A Cross-Cancer Genetic Association Analysis of the DNA Repair and DNA Damage Signaling Pathways for Lung, Ovary, Prostate, Breast, and Colorectal Cancer.

BACKGROUND DNA damage is an established mediator of carcinogenesis, although genome-wide association studies (GWAS) have identified few significant loci. This cross-cancer site, pooled analysis was performed to increase the power to detect common variants of DNA repair genes associated with cancer susceptibility. METHODS We conducted a cross-cancer analysis of 60,297 single nucleotide polymor...

Polymorphism of the DNA repair gene XRCC1 and risk of primary lung cancer.

DNA repair plays a critical role in protecting the genome of the cell from insults of cancer-causing agents, such as those found in tobacco smoke. Reduced DNA repair capacity, therefore, can increase the susceptibility to smoking-related cancers. Recently, three coding polymorphisms in X-ray cross-complementing group 1 (XRCC1) DNA repair gene have been identified, and it is possible that these ...

Polymorphisms of the DNA repair gene xeroderma pigmentosum group A and risk of primary lung cancer.

Polymorphisms in DNA repair genes may be associated with differences in the repair capacity of DNA damage and may influence an individual's susceptibility to smoking-related cancer. We investigated the association between two polymorphisms of the DNA repair gene XPA and risk of lung cancer in the Korean population. Two XPA polymorphisms (A23G and G709A) were typed in 265 lung cancer patients an...
